摘要:
目的:探讨miR-155对乳腺癌细胞增殖的影响。方法:选取乳腺癌患者20例,取患者乳腺癌及癌旁组织标本,使用实时荧光定量PCR技术检测乳腺癌、癌旁组织中以及MCF-7细胞中miR-155的表达,MCF-7细胞中Ki67的表达。应用CCK8法检测细胞的增殖能力。
结果:乳腺癌患者组织中miR-155相对表达量明显高于对应的癌旁组织。

乳腺癌是一种常见的恶性肿瘤,具有高发病率和死亡率。[1]找寻新型治疗乳腺癌的作用靶点并明晰其在乳腺癌当中的作用是当今的热点问题。
本研究聚焦乳腺癌这一重大疾病,以近期收治于本院的乳腺癌手术20例为临床样本,通过研究进而揭示miR-155在乳腺癌细胞增殖当中的作用.
1 材料与方法
1.1 组织样本
选择2018年6月至2018年9月期间来自河北工程大学附属医院乳腺癌患者,共计20例,取患者手术切除乳腺癌组织样本和癌旁组织样本,所有患者术前均未接受任何治疗。
1.2 方法步骤
乳腺癌MCF-7细胞系进行细胞培养,将合成的miR-155mimics、inhibitor和对应的NC进行转染,采用Trizol法进行全RNA 的提取、反转录、细胞增殖实验及数据处理。miR-155以U6为内参基因,Ki67以GAPDH为内参基因,表达量采用法进行基因表达量的计算。所有数据采用SPSS 20.0软件进行分析。数据采用平均数±标准差(x ±s)表示,采用t检验或单因素方差分析进行组间显著性分析,p <0.05被认为具有统计学意义。

3 讨论
miRNAs在乳腺癌的诊断和治疗中具有巨大的潜在应用价值[2, 3],miR-155与乳腺癌的发展过程具有强相关性[4]。本研究同样发现,miR-155的表达量在乳腺癌组织中显著升高。这些均提示miR-155为一种潜在的癌基因。
本研究以乳腺癌临床样本为依托,发现miR-155与细胞增殖相关蛋白Ki67的表达呈现正相关,进而在细胞水平上得以验证,最终发现在乳腺癌中miR-155作为一种micro RNA可通过调控Ki67的表达进而影响细胞增殖过程。

The role of miR-155 in the proliferation of breast cancer cells
Objective: To investigate the expression of miR-155 in patients with breast cancer (BC) .
Methods: A total of 20 patients with BC treated in our hospital were selected from June 2018 to September 2018. MCF-7 cells were cultured and divided into four groups: miR-155 mimic, NC mimic, miR-155 inhibitor and NC inhibitor. The expression of miR-155 and Ki67 in MCF-7 cells and breast cancer tissues were detected by qPCR, and the proliferation ability of MCF-7 cells was detected by CCK8 assay.
Results: The relative expression of miR-155 in breast cancer tissues was significantly higher than that in corresponding adjacent tissues.The expression of miR-155 was positively correlated with the positive rate of Ki-67 in breast cancer , the cell proliferation ability was also significantly inhibited .
Conclusion: miR-155 is highly expressed in breast cancer tissues..
